Species: Homo sapiens
Genetic Insert: Cyclin B1
Vector Backbone Description: Backbone Size:4500; Vector Backbone:pCMX; Vector Types:Mammalian Expression; Bacterial Resistance:Ampicillin
Defining Citation: PMID:10395539
Comments: The stop codon of cyclin B1 in the pCMX vector was mutated to a Mfe1 site to link cyclin B1 to MmGFP. Cyclin B1 was fused to the second amino acid residue of MmGFP by a 5 amino acid linker (AGAQF). The construct is cloned as a HindIII-BamHI fragment into pCMX.
The Cyclin insert contains a V5A polymorphism which has no functional consequence. More information on MmGFP can be found in the following reference: Zernicka-Goetz, M., Pines, J., M., Ryan, K., Siemering, K.R., Haseloff, J., and Gurdon, J.B. Genetic Insert: Tension Sensor module
Vector Backbone Description: Backbone Marker:Invitrogen; Backbone Size:5428; Vector Backbone:pcDNA3.1; Vector Types:Mammalian Expression; Bacterial Resistance:Ampicillin
Defining Citation: PMID:20613844
Comments: The are a few discrepancies with the author sequence. THese mismatches are in the pcDNA3.1(+) part of the vector, not the specific inserts. So they should be of little consequence.

Comments: Addgene's sequence with pBAD-F shows minor mismatches and gaps with the reference sequence from the depositing lab. These discrepancies are a result of mistakes made during sequence assembly and should not impact plasmid function.
Please note that growing the construct at 30-34C may increase protein yield (36-48hours).
