Species: Drosophila melanogaster
Genetic Insert: let-7 seed mutant perfect site
Vector Backbone Description: Backbone Marker:David Sabatini Lab; Backbone Size:0; Vector Backbone:pMT-puro; Vector Types:Insect Expression, Luciferase, Drosophila metallothionein gene promoter drives expression; Bacterial Resistance:Ampicillin
References:
Comments: Luciferase-reporter inserts were made by annealing oligonucleotides with their reverse complements, leaving overhangs for the indicated restriction sites (lower case): let-7-psm (gagctcACTATACAACCTACAAGCACAactagt). Annealed oligos were ligated into SacI/SpeI-cleaved pIS2. These plasmids were linearized with HindIII, polished with Klenow enzyme to create blunt ends, and digested with NotI to excise the Renilla luciferase gene with the modified UTR from the remainder of pIS2. The gel-purified Renilla gene fragment was then ligated into pMT-puro between EcoRV and NotI sites for copper-induced expression in S2 cells.

Genetic Insert:
Vector Backbone Description: Backbone Size:2686; Vector Backbone:pUC19; Vector Types:Mammalian Expression; Bacterial Resistance:Chloramphenicol and Ampicillin
References:
Comments: This plasmid was designed to tag transcripts of insert at the 3' with an 12X MS2 stem loop tag. Insertion of the transcript of interst is performed by BsaI cloning. 5' BsaI generates ACAG overhang (BsaI recognition site 2659-2664) 3' BsaI generates CCCG overhang (BsaI recognition site 4086-4091) These BsaI sites are lost upon insertion of the transcript of interest.
