The sequence is a consensus, however deviation from the consensus sequence ranging from 0.9 to 9.1% (mean deviation 3.9%) has been reported. The construct is mainly used as a FISH probe for visualising pericentromeric regions in mouse cells and this is unlikely to be affected by minor sequence variations. Another important point about this construct is that contains 8 direct repeats and the 234 bp satellite sequence also contains internal repeats. This will make it prone to rearrangements when it is grown up for isolation, which means that it will be difficult to completely guarantee its integrity.
