Comments: The insert (mPer-luciferase) derived from the pGL3 basic E2 vector (Plasmid #48747, a gift from Joseph Takahashi). The pLV6 backbone derived from the pLV6-Bmal-luc vector (Plasmid #68833, a gift from Steven Brown). The pLV6-Bmal-luc vector contains one BamHI cutting site 3’ of the Bmal1-luciferase insert. Site-directed mutagenesis was used to create an additional BamHI cutting site 5’ of the insert. The mPer2-luc insert of the pGL3-Per2-luc vector is by default flanked by BamHI cutting sites. After successful digestions and extractions, the pLV6 backbone and the mPer2-luc fragment were ligated with T4 DNA ligase, hence obtaining the pLV6-mPer2-luc vector.
