  • RRID:Addgene_24342

http://www.addgene.org/24342

Vector Backbone Description: Backbone Size:3000; Vector Backbone:pBluescript SK(+); Vector Types:Mammalian Expression; Bacterial Resistance:Ampicillin
Defining Citation: PMID:20434990
Comments: The BamHI/BglI fragment from pUAST (Brand and Perrimon, 1993), containing 5 copies of a GAL4 binding site and the Drosophila hsp70 minimal promoter was subcloned into BamHI site of pBluescript containing an optimized GFP split with an intron (GG) (Zong et al., 2005). This strategy generates a hybrid BglII/BamHI site in front of the GFP.

Genetic Insert: FLPo
Vector Backbone Description: Backbone Size:9000; Vector Backbone:pQUAST; Vector Types:Insect Expression; Bacterial Resistance:Ampicillin
Defining Citation: PMID:20434990
Comments: Mammalian codon-optimized FLP recombinase (FLPo; Addgene plasmid 13792) (Raymond and Soriano, 2007) was PCR amplified to include EcoRI/XbaI restriction sites, and cloned into pQUAS-DSCP to generate pQUAS-DSCP-FLPo.

Genetic Insert: LacZ
Vector Backbone Description: Backbone Size:9000; Vector Backbone:pQUAST; Vector Types:Insect Expression; Bacterial Resistance:Ampicillin
Defining Citation: PMID:20434990
Comments: The nuclear LacZ insert was PCR amplified from UAS-nucLacZ genomic flies (Bloomington Stock Center). The KpnI/XbaI digested nucLacZ PCR fragment was then ligated into pQUAST.

Defining Citation: PMID:19706417
Comments: ins-Cre_cryaa-Venus was generated by placing the Cre coding sequence downstream of 1.5 kb of the proximal insulin promoter. The cryaa-Venus cassette was inserted downstream of Cre in the reverse orientation using 0.54 kb of the cryaa promoter.

Proper citation: RRID:Addgene_24333 Copy

http://www.addgene.org/24334

Species: Danio rerio
Genetic Insert: H2B
Vector Backbone Description: Backbone Size:3000; Vector Backbone:pBluescript; Vector Types:Worm Expression, Cre/Lox; Bacterial Resistance:Ampicillin
Defining Citation: PMID:19706417
Comments: This plasmid was constructed using 1.5 kb of the hsp70l promoter. The floxed mCherry-STOP and H2b_GFP fragments were PCR amplified individually and sequentially cloned downstream of the hsp70l promoter. The mCherry-STOP cassette serves as a heat-shock induction marker prevents read-through translation of the H2B-GFP fusion protein, which is only expressed in cells that have undergone a Cre mediated excision event. cryaa-Cerulean was inserted downstream of HSB-GFP in the reverse orientation. *H2B has D26G and V119I mutations. According to the depositor, GFP expression from this construct was always nuclear so there is no reason to suspect that the mutations affect the function (nuclear localization) of the H2B fragment.
