  • RRID:Addgene_125620

http://www.addgene.org/125620

Species: Homo sapiens
Genetic Insert: PPAT
Vector Backbone Description: Backbone Marker:Clontech; Backbone Size:5472; Vector Backbone:pCI-neo; Vector Types:Mammalian Expression; Bacterial Resistance:Ampicillin

  • RRID:Addgene_125747

http://www.addgene.org/125747

Species: Synthetic
Genetic Insert: sfGFP
Vector Backbone Description: Vector Backbone:Custom; Vector Types:Mammalian Expression; Bacterial Resistance:Ampicillin
Defining Citation: PMID:32183906
Comments: Similar to the situation with some other plasmids in this collection, the missing four nucleotides at the 5' end of the WPRE renders it unrecognizable to SnapGene, but the expression vector is functional. Nevertheless, for sfGFP expression, the depositor recommends the updated versions of this plasmid, PB CAG-sfGFP (https://www.addgene.org/133568/) or PB-CA sfGFP (https://www.addgene.org/159094/), that can also be used without the piggyBac transposase. Red fluorescent protein versions are also available: PB CAG-tdTomato (https://www.addgene.org/133569/) or PB-CA tdTomato (https://www.addgene.org/159095/).

http://www.addgene.org/125748

Species: Synthetic
Genetic Insert: sfGFP-GSAx9-iCre-ERT2
Vector Backbone Description: Vector Backbone:Custom; Vector Types:Mammalian Expression, Cre/Lox; Bacterial Resistance:Ampicillin
Defining Citation: PMID:32183906
Comments: Linker between sfGFP and iCre-ERT2 is a clean GSAx9 sequence, similar in length to the mutated P2A sequence in the plasmid pCAG-sfGFP-iCre-ERT2 (#125749). The sfGFP fluorescence from the sfGFP-iCre-ERT2 is dim, as described in Nam and Capecchi, 2020 (https://doi.org/10.1186/s13064-020-00139-5) and a Figshare page (https://doi.org/10.6084/m9.figshare.13218437.v35). In short, it is not visible to the eyes, but can be detected with flow cytometry. Similar to the situation with some other plasmids in this collection, the missing four nucleotides at the 5' end of the WPRE renders it unrecognizable to SnapGene, but the expression vector is functional.
